چکیده

Partial hydrolysis of whey proteins by enzymes immobilized on an inert support can either change or evidence functional properties of the produced peptides, thereby increasing their applications. The hydrolysis of sweet cheese whey proteins by alcalase, which is multipoint-immobilized on agarose gel, is studied here. A MichaelisMenten model that takes into account competitive inhibition by the product was fitted to experimental data. The influence of pH on the kinetic parameters in the range 6.0 to 11.0 was assessed, at 50C. Initial reaction-rate assays in a pHstat at different concentrations of substrate were used to estimate kinetic and Michaelis-Menten parameters, k and KM. Experimental data from long-term batch assays were used to quantify the inhibition parameter, KI. The fitting of the model to the experimental data was accurate in the entire pH range.

A bench-scale, continuous vortex flow reactor (VFR), with a radius ratio, , equal to 0.48 and an aspect ratio, , equal to 11.19 was studied. This reactor may be used in the enzymatic hydrolysis of polypeptides obtained from sweet cheese whey with enzymes immobilized on agarose gel. Protein hydrolysis by immobilized and stabilized trypsin.

The preparation of novel immobilized and stabilized derivatives of trypsin is reported here. The new derivatives preserved 80% of the initial catalytic activity toward synthetic substrates [benzoyl-arginine p-nitroanilide (BAPNA)] and were 50,000-fold more thermally stable than the diluted soluble enzyme in the absence of autolysis. Trypsin was immobilized on highly activated glyoxyl-Sepharose ...

Effects of Enzymatic Hydrolysis on the Foaming Properties of Whey Proteins

Changes in the foaming properties of whey proteins concentrate (WPC) subjected to enzymatic hydrolysis were studied. Activity and stability of the foam generated from the enzymatic hydrolysis of WPC with two proteases: Trypsin from porcine pancreas (analytical quality) and Alcalase 2.4 L FG (Alcalase) (commercial food grade) were analyzed. WPC solution at concentration 100 mg/mL in 50 mM phosph...

In this study, immobilization of partially purified almond (Amygdalus communis) β-galactosidase on Con A layered calcium alginate-cellulose beads was investigated. Immobilized β-galactosidase retained 72% of theinitial activity after crosslinking by glutaraldehyde. Both soluble and immobilized enzyme exhibited the samepH and temperature optima at pH 5.5 and 50ºC, respectively. Howev...
